No report has previously clarified the origin of the surviving regional populations of Cottus pollux (Günther, 1873) in Shikoku. This study clarified the genetic relationships of the surviving C. pollux LE population (Kagawa population) in Shikoku and revealed that it possesses a unique haplotype. This Kagawa population, the only surviving C. pollux LE in Shikoku, showed the smallest genetic distance to the C. pollux ME population of the Takahashi River system in Okayama Prefecture. A haplotype network constructed using the genetic sequences of the Cottus species complex reported by Yamamoto (2018) and Kanno et al. (2018), retrieved from the International Nucleotide Sequence Database Collaboration (INSDC), placed the Kagawa population within the C. pollux ME clade. These findings suggest that the identification of the Kagawa population as C. pollux LE warrants further review.
